Sildenafil Citrate is a phosphodiesterase type 5 inhibitor used to treat erectile dysfunction and premature ejaculation in men. Sildenafil Citrate is a phosphodiesterase type 5 inhibitor which increases the blood flow to the penis to achieve and maintain an erection. Sildenafil Citrate is also known as tadalafil or tadalafil oral tablet, and is a phosphodiesterase type 5 inhibitor. In order to maintain an erection, the penis is relaxed. When sexual stimulation is aroused, nitric oxide is released and activates a chemical known as cyclic guanosine monophosphate (cGMP). This chemical relaxes the smooth muscle in the corpus cavernosum and causes an erection. Sildenafil Citrate is used as a phosphodiesterase type 5 inhibitor, and is available in several forms, including tablets and capsules. The dosage of sildenafil citrate is 100 mg (60 tablets) or 100 mg (30 tablets) and is taken by mouth once a day. It is important to follow the instructions given by your healthcare provider for safe usage of this drug.
Dosage and direction: Take one tablet of sildenafil citrate orally with a glass of water 30 minutes before sexual activity. Do not take more than one tablet in a 24-hour period. Swallow the tablet whole with a full glass of water. Take the tablet at least 30 minutes before sexual activity. If you have taken too much, it may cause an overdose. The overdose may be life-threatening and require prompt medical attention. If you have taken sildenafil citrate, do not take it more than once in 24 hours. If you think you have taken too much, it may be dangerous and require prompt medical attention. Sildenafil Citrate should not be taken more than once in a 24-hour period. Avoid alcohol consumption while taking sildenafil citrate. Sildenafil citrate may interact with nitrates, medicines used to treat angina, medicines used to treat high blood pressure, and medicines used to treat pulmonary arterial hypertension (high blood pressure in the lungs).

Chloramphenicol

Acetazolamide
Therapy should be administered with caution because the use of Sildenafil increases the effect of acetazolamide by pharmacodynamic synergism and it also increases the risk of hypotension.
The use of this medicine can lower blood pressure and when combining it with alcohol further increases the effect. You may experience dizziness, lightheadedness, fainting, flushing, headache, and heart palpitation. Also, avoid consuming grapefruit juice because it may increase the effect of avanafil on blood levels.
Cardiovascular disease:
The use of phosphodiesterase-5 enzyme inhibitors is contraindicated in patients with cardiovascular disease. A doctor should consider the vasodilatory effects of this drug and whether they may affect patients with cardiovascular disease. The use of this drug may increase the risk of angina pectoris, AV block, myocardial infarction, ventricular arrhythmia, tachycardia, palpitation, hypotension, postural hypotension, syncope, cerebral thrombosis, cerebrovascular hemorrhage, transient ischemic attack, cardiac arrest, heart failure, and hypertension. These events can occur during or after sexual activity.
Renal dysfunction:
Therapy should be administered with caution in patients with severe renal disease or on renal dialysis. The plasma clearance of this drug is decreased in patients with a severe renal impairment which results in drug accumulation.
Liver diseases:
The use of this drug is not recommended for patients with severe hepatic impairment because the pharmacokinetic disposition of this drug has not been assessed in patients with severe hepatic impairment.
What is liver disease? and How do I know?Liver diseasesare glandular degeneration caused by high levels of a tumor necrosis factor a or by excessive production of a prostaglandin E2 enzyme. This enzyme is associated with the synthesis of nitric oxide and cyclic adenosine monophosphate. The nitric oxide then activates the guanylate cyclase of the liver. When this enzyme is activated, the monophosphateukemia-NO is released in the aortic muscle causing inflow of blood into the aorta and outflow of the aortic muscle. The monophosphateukemia-NO then inhibits the phosphodiesterase enzyme which leads to degradation of cyclic adenosine monophosphate in the liver. This leads to accumulation of cyclic adenosine monophosphate in the aortic muscle and a decrease in blood pressure.Phosphodiesterase inhibitors are classified as L-type Ca2+ channels in that they have more than one high-voltage open in the aortic and that they have more than one low-voltage open in the mitral root. They have both low and high-voltage current densities.
Phosphodiesterase inhibitors are usually started at low dosages and gradually increased to a maximum recommended Dosis level of at least 2 mm.
